Von Dezember 1847 bis Februar 1853 lebte und arbeitete er in Sierra Leone, dem in Westafrika errichteten britischen Protektorat für befreite Sklaven. Koelle lehrte am Fourah Bay College, das 1827 von der Church Missionary Society gegründet wurde. 1859 wurde er von der Church Missionary Society nach Konstantinopel (Istanbul) entsandt, er blieb dort bis 1879.
