Er studierte klassische Philologie und orientalische Sprachen (arabisch, türkisch und persisch) in Heidelberg, Wien und Istanbul (Konstantinopel). 1815 bereiste von Richter mit dem schwedischen Diplomaten und Geistlichen Sven Fredrik Lidman (1784-1845) Ägypten und Unter-Nubien; 1815-1816 aber Palästina, Syrien, Zypern, Makedonien, Istanbul und Klein-Asien. Auf dieser letzten Reise erlag er einer Infektionskrankheit.
